Research Abstract |
We studied the clinical and immunolgical characteristics of 83 Zombian patients with Kaposis sarcoma(KS)and collected biopsy samples from some of these patients after getting their informed consent. We found thet some of the KS patients still maintained high CD4 counts, but those with an oral lesion(s)had significantly lower CD4 counts. We are now determining the vMIPs nucleotide sequences from biopsy samples for comparison with those of laboratory HHV-8 strains. To analyze the effects of chemokine-like proteins encoded by the human herpesvirus type 8 on HIV infection, we amplified and isolated viral MIP(vMIP)-I, -II, and -III genes of human herpesvirus type 8(HHV-8)from BCBL-l cell line by PCR.We then constracted these genes into an expression vector and transfected these vectors into a 293T cell line. Culture supernatant containing secreted vMIPs showed the activity of Ca2+ influx to THP-1 cell line. vMIPs genes were also introduced into Cos 7 cells and permanent cell lines secreting vMIPs have been established. We purified a large amount of vMIPs and are studying the effects of these prroteins on HIV infection and immunological effects. Similar experiments are being done using the vMIPs genes of HHV-8 isolated from KS patients.
