Research Abstract |
1. Subjects and Methods : Cytokines production of peripheral blood mononuclear cells (PBMCs) stimulated with phorbol 12-myristate 13 acetate (PMA) ad ionomycin was investigated in atopic dermatitis patients with hen egg hypersensitivity. Further, the effects of thophylline on cytokines production of PBMCs in atopic dermatitis patients with hen egg hypersensitivity. Cytokines were measured in intracellular IFN-γ or IL-4 production by flow cytometry. 2. Results : (1) IFN-γ or IL-4 production of CD4+ T cells in atopic dermatitis patients with hen egg hypersensitivity. There were no differences of IFN-γ production in CD4+ T cells between patients with hen egg hypersensitivity and healthy children. In contrast, the frequency of IL-4-producing CD4+ T cells from the patients was significantly higher than that from healthy control subjects. producing CD4+ T cells did not change. (2) Effects of theophylline on IFN-γ or IL-4 production of CD4+ T cells IFN-γ production cells of CD4+ T cells was 9.58%, and that in theophylline-stimulated PBMCs increased to 12.40%. IL-4 production cells of CD4+ T cells was 1.60%, and that in theophylline-stimulated PBMCs did not change, 1.64%. IFN-γ production of CD4+ cells was significantly increased in theophylline-stimulated cultures, however, there were no changes in IL-4 production of CD4+ T cells in spite of theophylline added. (3) Effects of theophyllinc on IFN-γ or IL-4 production of CD8+ T cells IFN-γ production cells of CD8+ T cells was 26.94%, and that in theophylline-stimulated PBMCs decreased to 12.40%, but there were no significant differences. IL-4 production cells of CD8+ T cells was 6.58%, and that in theophylline-stimulated PBMCs did not significantly decrease, 4.05%. There were no changes in IFN-γ or IL-4 productions of CD8+ cells theophylline-stimulated cultures. 3. Conclusion : IFN-γ production of CD4+ T cells was increased by theophylline, however, theophylline did not influence on IL-4 production of CD4+ T cells. On the other hand, IL-4 production of CD8+ T cells tended to decrease by theophylline. From these results, although theophylline is an anti-inflammation drug for bronchial asthma, it might be a drug for atopic dermatitis with food hypersensitivity in which influence on the TH1/TH2 imbalance. 4.
