Research Abstract

Plasma concentralion of GLP-2, histological changes of intestinal mucosa were investigated in rats with ileo-Jejunal Transposition(IJT)that show hyperenteroglucagonemia. Then, the efferct of external administration of glicenlin, GLP-1, and GLP-2 were evaluated regarding to the trophic effect on intestinal mucosa both in vitro and In vivo. The results were obtained as fonows 1)Plasma levels of GLP-2 in IJT rats were significantly higher than control rats showing the marked increase of villous heights of jeuno-ileal mucosa. 2)Extrnal administralion of GLP-2 significantly enhances the ODC activity in both jejuno-ielal mucosa in normal rats, which indicates the early phase trophic effect. Levels of GLP-2 were almost same as the levels of those in rats with IJT.3)Both the ODC activities and tritium thymidine incorporation of IEC-6 were significantly stimulated by administration of GLP-2, which produces higher concentration of GLP-2 in the medium than the plasma concentrations of IJT rats. 4)Glicentin has the similar effect as shown in the experiments performed by the using GLP-2, however those effects of glicentin were less significant than GLP-2. 5)GLP-1 has no stimulating effects on parameters as described above both in vitro and vivo.
These results indicate a possible application of GLP-2 to the treatment of intestinal ulcer healing in patients with Crohn's disease. Moreover, it may be a new strategy to use of IJT in the surgical treatment of Crohn's disease.
