Research Abstract

a. Establishment of high-metastasis cell lines : Cell lines that have the high ability to metastasize to the liver and lung using MH134 (hepatoma cell line of C3H/HEN mice) were established.
b. Confirmation of the ability of metastasis and tumor formation in these cell lines : The ability of metastasis and tumor formation in these cell lines by injecting to C3H/HEN mice was confirmed. All these cell lines metastasized and formed solid tumors macroscopically in the liver and lung.
c. The isolation of mRNA of these cell lines and search of specific genes responsible for high metastasis to the liver and/or lung : Using isolated RNAs, those specific genes are being investigated by the subtraction method. Similar analysis began to be performed by the DNA microarray method.
d. Electroporation-mediated transfection of genes : Electroporation-mediated IL-12 gene therapy for hepatocellular carcinoma was found to be effective for both main tumor and distant metastasis in the mice model (Cancer Research, in press).
e. Discovery of the importance of deacetylation on metastasis in hepatocellular carcinoma : The overexpression of histone deacetylase 1 was found to have a strong correlation with the dedifferentiation and metastasis of hepatocellular carcinoma.(Hepatology, submitted)
