Research Abstract

The insulin-like growth factor (IGF) system, which is composed of two ligands (IGF- I, IGF- II), two receptors (IGFR- I, IGFR- II) and six binding proteins (IGFBP- 1 to - 6), plays an important role in cell biology. The mRNA expression of the IGF system in human prostate cancer tissue was examined. Expression of the IGF system, except IGFBP- 1, was detected in all prostate cancer tissue samples. The expression of IGF- II and IGFBP- 2 significantly correlated with pathologic stage, lymph node metastasis, histologic differentiation and PSA levels after hormone therapy. However, expression of IGF- I significantly lower in advanced patients. In conclusion, in the IGF system, IGF- II and IGFBP- 2 play a role in human prostate cancer progression and their increased expression is a possible candidate for a prognostic indicator in hormone-treated prostate cancer patients.
IGFBP- 7 mRNA, which include in IGFBP superfamily, were expressed in the human BPH derived epithelial and stromal cells by RT-PCR method. However, the expression of IGFBP- 7 mRNA was not detected in human prostate cancer cell line. The results of Northern blot analysis showed the absence of expression in prostate cancer tissues. The inhibitory activity of IGFBP- 7 on cell growth rate might be applicable for the treatment of patients with prostate cancer.
