Research Abstract |
Nasal mast cells(NMC) in allergic rhinitis patients exhibit increased expression of the FcεRI, and TL-4 as well as IgE can upregulate the FcεRI expression in mast cells (J Clin Invest 1997,Clin exp Allergy 1998). This enhanced expression of the FcεRI in NMC was associated with an increase in IgE-bindability as well as increased mediator release. Furthermore, the FcεRI expression in NMC correlated well with the levels of serum IgE.. Moreover mast cells can induce IgE synthesis in B cedlls. Gamma delta T cells are also increased in allergic rhinitis and they release higher levels of IL-4 and IL-13 than alpha beta T cells. Here we first analyzed the local IgE synthesis in the nasal mucosa of patients with alletgic rhinitis after invitro stimulation with Derf II. (mite antigen) by RT-PCR. We then analyzed the effect of DEP on cytokine production (IL-4,IL-5,IL-6 and IL-13) from mast cells and gamma delta T cells by measuring the levels in the culture supernatants after stu=imulating with DerfII alone or DerfII in combination wit DEP. An upregulation of the c epsilon mRNA was detected in vitro after stimulating with Derf II. IL-4 and IL-13 mRNA were also upregulated. DEP produced more enhanced cytokine secretion from nasal mast cells as well as gamma delta T cells. These findings suggest that local IgE synthesis occurs at sites of allergic inflammation and indicates a very important role for NMC in regulating chronic allergic inflammation by inducing local IgE synthesis. DEP has a key role in enhancing allergic inflammation by its effect on mast cells and gamma delta T cells also.
