Research Abstract |
The present study has been designed to analyze of the roles of several kinds of growth factors in the morphogenesis of the gustatory and non-gustatory papillae of the tongue. For this purpose, three different methods have been employed ; ultrastructural observations of gustatory and non-gustatory papillae of the tongue of rodents, and immunohistochemistry and in situ hybridization of keratin subunits in the lingual epithelium and of growth factors in the lingual mucosa in rats. The results obtained as follows ; 1. Ultrastructural observations of the lingual epithelium of rats and mice showed that the period of formation of gustatory and non-gustatory papillae was clearly different. The rudiments of fungiform papillae were recognized prior to formation of those of filiform papillae. 2. Keratinization of the lingual epithelium progressed with the advance of morphogenesis of filiform papillae, and started just before birth. 3. ryo of rats and on the 15th day of embryo of mice by the depression of the epithelium into the deep area in the connective tissue prior to the approach of the sensory nerve to the dorsal epithelium of the tongue. These rudiments moved to the dorsal epithelium from both sides of the median sulcus to the lateral sides. The differentiation of taste buds had not begun in this stage. 4. Immuno-reactivity of polyclonal antibody of keratin and monoclonal antibody of keratin subunits was negative in the dorsal epithelium of the tongue of the fetuses of rats. That of polyclonal antibody of keratin widely recognizable in the lingual epithelium of rats during the postnatal development of rats. Among monoclinal antibody of keratin subunits, keratin 14 showed a positively immuno-reactivity in the basal layer of the lingual epithelium during the whole period of postnatal development. Slight reactivity of keratins 13 and 19 has been recognized in the restricted area of the lingual epithelium during the same period. 5.
