Research Abstract |
1) Isolation of chick lefty cDNA In order to isolate a chick lefty cDNA, we synthesized degenerated two oligo DNAs based upon the conserved amino acids sequences between mouse and human. Using the primers, we performed RT-PCR : template was total RNA which was prepared from early chick embryos. We isolated several cDNA clones. Among them one clone was highly homologous to mouse and human lefty cDNAs. We studied its expression pattern during early chick development using by whole-mount in situ hybridization. We found that lefty1 is expressed asymmetrically on the left side of the prospective floor plate, notochord and lateral plate mesoderm of the chick embryo. 2) Identification of the genes which plays a role in establishment of left-right axis and an analysis of the expression patterns To isolate unknown genes involving left-right axis, we performed subtraction screening using with the cDNAs separately prepared from left and right sided bodies. We isolated a cSnR which encode for a transcriptional factor that had a zinc finger domain. Since cSnR was expressed in right lateral palate mesoderm in the early chick embryo as published previously, we performed the expression patterns in the late chick embryo. We found that cSnR was expressed in the genital ridge of 5 days embryo in both side and in both sexes. At 7 days embryo cSnR was expressed at the right side of the genital ridge in the male but was not expressed in the female. These results suggests that cSnR was involed in the morphogenesis of the genital ridge
