Research Abstract

DNAメチル化によって発現が制御されヒトがんの生物学的特性の決定に寄与する遺伝子の単離を試みるため、DNAメチル化阻害剤である5-azacytidine処理により増殖が抑制された培養胃がん細胞株においてdifferential display法を施行した。5-azacytidine処理によりWip1,TROP2,B4-2,BRCA-2 region transcription unit CG005,cofilin,ROCK,CD44ならびにinterferon-inducible protein 6-16 mRNA発現の経時的な変化を認め、5-azacytidine処理によって発現の変化する未知遺伝子も単離し得た。これらの遺伝子のmRNA発現を臨床手術材料においても検討したところ、非がん粘膜に比し胃がんにおいてWip1,B4-2ならびにcofilinの、非がん粘膜に比し大腸がんにおいてWip1,B4-2,BRCA-2 region transcription unit CG005,cofilinならびにROCKの、非がん肝組織に比し肝細胞がんにおいてTROP2,B4-2ならびにBRCA-2 region transcription unit CG005のmRNA発現の有意な低下を認め、大腸がんにおいてTROP2発現は有意に亢進していた。Wip1ならびにBRCA-2 region transcription unit CG005の発現低下は肝細胞がんの分化度と有意に相関し、門脈侵襲を伴う肝細胞がんにおいて特に発現が低下していた。TROP2発現は正常肝組織に比し前がん状態にある慢性傷害肝において有意に亢進し、肝細胞がんの悪性進展に伴って逆に低下していた。B4-2の発現は特にB型肝炎ウイルス感染に伴って低下していた。これらの知見は、DNAメチル化がこれらのがん関連遺伝子の発現を直接または間接に制御してヒトの多段階発がんに寄与する可能性を示している。
