Research Abstract

毒素原性大腸菌の産生する耐熱性エンテロトキシン(STa)は、腸管上皮細胞膜上に存在する受容体蛋白質(STaR/GC-C)に特異的に結合して、ヒトや家畜に急性の下痢を引き起こす毒素である。STaRの構造と活性の相関を明らかにするには、動物細胞に発現させたSTaRは蛋白質の発現量が少なく、あまり蛋白量を分取できない。また、STaRを大腸菌に発現させると、蛋白量は分取できるが、生物活性を失っていた。そこで、バキュロウイルスを用いて昆虫細胞にSTaRを発現させたところ、大量の蛋白質の発現が認められ、生物活性も認められた。リコンビナントSTaRは、STa結合活性(Kd=62nM)を示し、グアニル酸シクラーゼ活性も、動物細胞に発現させたSTaRと同じ生物活性を示した。次に、STaRの細胞外領域の部分蛋白質を、昆虫細胞に大量に発現させた。光親和性官能基を持つSTaアナログを用いて、分取したSTaRとインキュベーションして光架橋をおこない、Lys-Cにて酵素消化して、STaの結合している領域を質量分析器にて解析した結果、387残基めから393残基めの部分に結合していることがわかった。この結果は、STaRの細胞膜に近い部分でSTaが結合していることが示唆された。
