Research Abstract

HIV-1感染無症候キャリアー(AC)の末梢血CD8陽性T細胞はHIV-1抑制活性を有することが知られており、この抑制には複数の機序が関与していると考えられている。これまでにこの活性がMHC-I拘束性と非拘束性のものに分けられることやMHC-I拘束性の主な活性がCTLによる細胞傷害作用であることが判明しているが、非拘束性の活性の詳細は不明である。我々の研究からは、HIV-1感染者の病態が進行していく過程において、MHC-I非拘束性の抑制活性が拘束性の抑制活性よりも先に消失していく傾向が認められ、HIV-1抑制活性のバランスの破綻が病態の進行に関わっている可能性も示されており、この抑制のメカニズムの解明は、発症予防を考える上で重要な課題といえる。本研究では、Green Fluorescent Protein(GFP)遺伝子がHIV-1遺伝子に組み込まれた複製可能ウイルス(HIV-1.GFP)を用いてMHC-I非拘束性抑制活性に関与する因子の作用機序の解明を試みた。
はじめに、HIV-1.GFPをMOLT-4とkit225細胞に感染させ、MOLT-4/HIV-1.GFPとkit225/HIV-1.GFP細胞を樹立し、これらの感染細胞株の抗HIV-1活性感受性を検討した。その結果、AC由来CD8陽性T細胞をkit225細胞と共培養することにより、kit225細胞の蛍光強度を減弱させる活性、すなわちHIV-1抑制活性が検出されることがFACSによる解析から確認された。この活性はAC由来CD8陽性T細胞のなかの約67%で検出され、GFPの発現が低下した細胞においてもアポトーシスは認められなかった。すなわち、kit225/HIV-1.GFP細胞においてはMHC-I非拘束性の細胞傷害を伴わない抑制活性が検出可能であることが示され、この活性の詳細な解析に有用であることが判明した。
