Research Abstract

初年度は,CGH法およびSKY法を用いて神経膠芽腫細胞株の染色体数的異常,構造異常を解析し,増幅領域は高頻度に,転座,挿入などの構造異常を示すことが明らかになった.今年度はさらに症例を重ね検討し,YAC cloneを使用してfluorescence in situ hybridization(FISH)により異常領域を詳細に特定した.増幅のある領域でtranslocationやinsertionがの91.7%と極めて高頻度に認められた。またこれらはHSRやDMとは異なり4-10倍程度の増幅であった。悪性グリオーマでは異なる細胞株に7種類の共通した染色体構造異常が認められた。神経膠腫の臨床検体を用いたCGH解析の臨床的有用性については、本年度はさらに症例を重ね検討を行なった。8番染色体ではPCR-SSCP法によるp53遺伝子異常と相関し、7番染色体短腕の増幅はEGFR遺伝子増幅と相関した。また8番染色体を有する症例は7番染色体短腕の増幅症例に比べ、若年者に多く予後良好であった。以上のごとくCGH法の臨床的有用性について統計学的に立証しえた.腫瘍内heterogeneityについては,7番,10番,11番,20番染色体の特異的プローブを用いたFISH法とlaser scanning cytometerにより評価した。染色体不安定性はDNA aneuploidyの発現と密接な関係があり,神経膠腫で7番染色体不安定性が悪性神経膠腫患者の予後に関与することが明らかになった。
