Research Abstract

アレルギー性結膜炎は先進国では全人口の20%以上に認められ、重篤な場合は角膜傷害に至る。本年度の目的は、ヒトアレルギー性結膜炎モデル(実験的眼瞼結膜炎:EC)発症におけるサイトカインの役割を明らかにすることとした。ブラウンノルウェー(BN)ラットに抗原としてラグウィード(Ragweed,RW)を用いて能動免疫、受動免疫によりECを誘導し(Graefe's.Arch.Clin.Exp.Ophthalmol.238:346,2000)、結膜炎は病理学的に、リンパ節は免疫学的に解析した。能動免疫の際はIL-12を腹腔内投与、受動免疫の際は抗原に加えてIL-4あるいはIL-12を添加培養後BNラットに移入した。これらのラットにRWを点眼しECを誘導した。IL-4を添加することによりTh2型免疫応答と好酸球浸潤の増強、IL-12の添加あるいは投与によりTh1型免疫応答、リンパ球浸潤の増強を認めた。以上の結果よりアレルギー性結膜炎病像形成におけるIL-4の重要性が判明した。さらにTh1誘導サイトカインであるIL-12の投与では病気は抑制できず、逆に病像の異なる強い病気が誘導されることが判った(Clin.Exp.Immunol.122:28,2000)。
また、アレルギー性結膜炎発症における内在性サイトカインの役割を検討するため、IL-4、IL-12、IFN-gノックアウトマウスに同様なシステムで結膜炎を誘導した。その結果、IL-4は結膜炎の発症に必須で、IFN-gは発症抑制に働いていると考えられた。興味あることに、遅発相の誘導にはIL-12も重要であることが判明した(J.Allergy Clin.Immunol.105:299,2000)。
