Research Abstract

A)我々が単離した象牙質特異的な遺伝子デンチンシアロプロティン(DSP)の転写制御領域の下流にBMP2,4の受容体であるActivin like kinase3(ALK3)のキナーゼ領域を取り除いたドミナントネガテブ型のALK3を連結したトランスジェニックマウスを作製した。独立な4系統のトランスジェニックマウスのうち3系統のマウスに歯牙に特異的に導入遺伝子の発現を認めた。しかしながら、歯の形態には明らかな異常を認めなかった。この転写制御領域により歯の発生の後期に象牙質特異的に遺伝子を発現させる事ができるが、上記の結果から歯の発生後期には象牙芽細胞にはALK3を介したシグナルは無関係かあるいは、発現量が弱く十分なドミナントネガテブ効果が得られていない可能性が示唆された。
B)Dsp-LacZマウスを用いて歯の分化誘導系を確立するために、まず歯胚培養法を用いて実際に象牙質でのみレポーター遺伝子のLacZが発現しているかを調べた。生体内同様に、培養歯牙にても象牙質特異的にLacZの発現が誘導された。現在、この方法を用いてALK3が象牙質の分化に深く関与するか否かを解析するために、我々が単離した制御領域を用いる場合よりも早い時期からALK3に対する阻害を行い、歯の分化に影響を与えるか検討している。
