Research Abstract

口腔腫瘍に対する放射線治療に起因する骨障害の病態解明を、骨芽細胞におけるアポトーシス機能を解明する方面から追求してきた。これまでに骨芽細胞様細胞にアポトーシスが生じ、そのpathwayの一部に蛋白質リン酸化及び脱リン酸化反応が関与していること(Morimoto Y.et al.1997)、蛋白合成とRNA合成が必要であること(Morimoto H.et al.1999)及びその蛋白の一つはFas抗原/リガンドであることを発見した(Morimoto Y.et al.2000)。平成11年度の研究期間中に、アポトーシス細胞で鍍銀染色に陽性反応を示す核小体形成帯いわゆるAgNORsが消失すること、アポトーシス誘導の際に、鍍銀染色に陽性反応を示す110kDa核内蛋白が消失し、80kDaの鍍銀染色に陽性反応を示す蛋白が出現することをcell-free systemを用いて発見した(Morimoto Y.et al.in press)。同時に、110kDaの蛋白の変化はアポトーシスの実行機構であるDNA fragmentaionと密接な関係があることも発見した。さらに、オカダ酸誘導アポトーシスの一部には蛋白分解酵素の一つであるcaspase3及びZn^<2+>の存在下で活性化するDNAエンドヌクレアーゼが関与していることを発見した(Morimoto Y.et al.in preparation)。
