Research Abstract

1)新規五座鉄錯体,即ち,種々のニトリル(ベンゾニトリル,1-フルオロベンゾニトリル,3,4-ジフルオロベンゾニトリル,ベンタフルオロべンゾニトリル)から由来する側鎖を第五配位子する[2,4-bis(2-pyridy lmethy limino)pentane]鉄(II)錯体の誘導体を合成し,過酸化水素存在下,pUCl9プラスミドDNAのDAN切断反応に及ぼすフッ素置換基効果について検討した.一定時間後DNAを電気泳動法にによってForm I,Form II,Form IIIの生成比を求めた.フッ素の数の増加と共にDNA切断活性能が向上しペンタフルオロベンゾニトリルを置換した錯体(1)において最も活性が高いことがわかった.また,種々のヒドロキシルラジカルスカベンジャー共存下においても切断反応は抑制されないことがわかった.錯体(1)のアセトニトリル溶液に過酸化水素を添加し,室温で可視吸収を測定すると587nmに極大吸収を持つ化学種が生成し,これは77KでのESRスペクトルでg値2.14,2.08,1.99を与え鉄(III)ヒドロペルオキソ種であることが判明した.以上の結果から錯体中のイミノ基の水素とヒドロペルオキシドの酸素間での水素結合を通して鉄-ヒドロペルオキシド中間体の0-0結合の不均等開裂が容易に起るためだと推定した.
2)三脚型配位子,N-(4-nitro-2-hydroxy)phenylmethyl-N-(2-pyridylethyl)-N-(2-pyridylmethyl)amine(HNE),硝酸第二鉄・9水和物をメタノール中で混合し鉄(III)錯体を単離した.X線結晶構造解析から,HNEは5座配位子として鉄に配位し,さらに二つのヒドロキソイオンが2つの鉄を架橋したビス(μ-ヒドロキソ)複核鉄(III)構造であることがわかった.この構造は,これまでに数例しか報告されておらず稀な構造である.単離した複核鉄(III)錯体-過酸化水素系によるシクロヘキサンの酸化反応を検討したところ,シクロヘキサノールへのヒドロキシル化が円滑に進行することがわかった.
