Research Abstract

昨年度は多波長低コヒーレンス光干渉を用いて表皮下末梢血管系の酸素濃度分布を測定し、高分解能光機能イメージングを行った。この結果を踏まえて、本年度は<10μmの空間分解能で末梢血管系の血流分布を測定・イメージングして、生体機能情報を得る基礎実験を行った。
(1)低コヒーレンス干渉光学系を単一モード光ファイバカップラーで構成し、測定系の小型・安定化を行った。本装置を用いて、ヌードマウス耳の表皮下末梢血管系のOCT像をin vivoで取得して、血管の微細構造を<10μmの空間分解能で可視化した。
(2)生体組織中での本手法の光到達深度の向上と空間分解能の定量化のために、照射レンズの集光効果を検討した。この結果、波長0.8μm帯において到達深度が約1mmから約3mmまで向上した。さらに、生体深部の空間分解能も<10μmが得られた。
(3)これまでの生体組織の屈折率と厚さ同時測定技術を併用して、新たに実サイズでの生体光トモグラフィを考案した。本手法を用いて、爪板や表皮下組織などの多層構造の可視化を行った。
(4)光散乱が少なくなる長波長帯1300nmの低コヒーレンス光源を用いて、生体組織構造の可視化を試み、0.8μm帯の約2倍の到達深度が得られた。
(5)レーザドップラー血流計測と低コヒーレンス干渉計測とを組み合わせて、毛細血管系の血流分布を分解能〜10μmでのイメージングを検討した。
(6)信号処理系の雑音の低減とデータ取り込み時間の短縮を行い、さらに微弱光検出系を整備して、深部の生体機能イメージングの基礎実験を行った。
