Research Abstract |
1. Postprandial suppression of blood glucose level (BGL) by anthocyanins. In order to optimize the postprandial BGL by food components for the prophylaxis of diabetes, various plants rich in authocyanins were investigated with respect to α-glucosidase (AGH) inhibition. As a result of in vitro screening on AGH inhibition, Ipomoea batatas cv Yamagawamurasaki purple sweet potato (YGM) had a potent maltase inhibition with the IC50 value of 0. 26 mg/ml. The YGM extract also showed a potent in vivo supressive effect of BGL after single oral administration of maltose in SD rats. As a result of isolation of active compound in YGM, acylated anthocyanins were found to show a potent AGH (maltase) inhibitory activity. 2. iminstration of IVY (5mg/kg), a significant (P<0. 05) blood pressure (BP) reduction was observed at 8 min (ΔMBP=19. 2 mmHg), and the reduction was held for 20 min. By using rat and human plasma. IVY was found to be metabolized by the action of aminopeptidases in plasma to form a subsequent ACE inhibitor with hypotensive effect, VY. Thus, the intake of IVY or wheat germ hydrolysate would serve in reguating BP by the combined depressor effect of IVY and VY, after absorption. 3. latent antihypertensive effect of buckwheat (BW) protein. Latent production of ACE inhibitors from tartary BW, and the characterization of peptides responsible fcr ACE inhibition were investigated. Sequential digestion with pepsin, trypsin and chymotrypsin resulted in a significant increase of ACE inhibitory activity (IC50 ; 0. 14 mg-protein/ml). rutin did not show the ACE inhibition. About 30 mmHg of BP reduction in SHR was observed after single oral adiministration of BW digest at 4 h. From the BW digest, 11 peptides were identified as an ACE inhibitor.
