磁力は骨芽細胞の分化・増殖を著しく亢進させる

Research Abstract

平成11年度において、微小磁性体(0.05mm直径)を導入したマウス頭蓋冠骨芽細胞(MC3T3-E1株)に対し、永久磁石(希土類磁石)を用いて細胞の水平方向に正磁場をかけたところ、その骨芽細胞(骨細胞に分化する)が両極方向に伸展することを観察した。その際、対照として、(1)磁性体を導入しないで磁場だけをかけたもの、(2)磁場をかけず、磁性体だけを導入しただけのもの、(3)磁性体も導入せず、磁場もかけないもの等について、細胞の増殖・分化度を顕微鏡レベルで解析した。その際、磁性体を導入し、かつ、磁場をかけた場合、上記対照細胞[(1)〜(3)]に比べ、細胞の結節形成能が早まることを見い出した。
そこで、平成12年度は、特に、ヒト骨芽細胞(normal human osteoblast cell system)を用いて、機械的伸張と磁場が細胞分化に及ぼす影響を観察し,かつ、分化にかかわる細胞内シグナル伝達系としてのMAPKカスケードについて調べた。その結果、、(1)〜(3)に示すコントロール群の骨芽細胞は、培養開始後12日後に最初の結節が現れ、25日で石灰化を検出した。ところが、磁性体を導入し、かつ、磁場をかけたものは、8日に最初の結節が観察され、18日目には石灰化が検出され,以後、石灰化が増加した。
また、磁性体を導入し、かつ、磁場をかけた骨芽細胞では、最終分化マーカーであるオステオカルシンの発現量が上昇し、MAPKカスケードに関しては、p38MAPKのみ活性が上がり、p44(Erk1),p42(Erk2)およびJNK/SAPKには変化がなかった.このように、本研究の成果として、細胞の進展のような、機械的刺激に対してはp38MAPKが特異的に応答する可能性が示唆された。
