Research Abstract

単離した肝細胞集団の中から、多能性を持つ肝幹細胞をFACS(Fluorescence activated cell sorter)とモノクローナル抗体を用いた細胞分離法により濃縮・純化し、それらの機能解析を行うことを目的として研究を行った。昨年度までの研究により、c-Kit-CD49f+CD29+ CD45-TER119-細胞中に限定して増殖能の高いコロニー形成能を有した細胞が存在し、肝細胞と胆管細胞という異なる細胞系列へと分化し得ることを明らかにした(Hepatology 32(6):1230-1239,2000.)。今年度は、c-Kit-CD49f+CD29+CD45-TER119-細胞の画分化を段階的にすすめ、自己増殖能をも備えた幹細胞画分を明らかにすることを試みた。その結果、c-Met+c-Kit-CD49f+/lowCD29+CD45-TER119-細胞中に限定して増殖能、多分化能、自己複製能、組織再構築能を兼ね備えた肝幹細胞(hepatic stem cell)が高頻度に存在することが判明した。また、この肝幹細胞の少なくとも増殖にhepatocyte growth factor(HGF)が必須の因子であることを明らかにした。今後、さらに、純化した肝幹細胞の分化・増殖・自己複製機構を培養系を用いて解析し、「臓器再生」の基盤となる知見および技術を確立する予定である。
