Research Abstract

本研究計画では、酵母(S.cerevisiae)内にて、広く哺乳動物の転写因子活性化能の定量を可能とするシステム(yeast-based luciferase assay;以下YLAと略)の構築を目的とする。それにより、転写因子の制御因子を単一あるいは複数個同時発現して制御システムを酵母内に再構築して検討を可能とする。本年度は以下の成果を得た.
1)p53 responsive element(p21,Bax)を含むプロモータ駆動をFirefly luciferaseをリポーターとして測定するシステムを完成,Renilla luciferase(内部標準)とともにp53,p73,p63によるプロモーター活性化の測定に成功した.
2)p53の変異体80種の転写活性化能を測定し,変異体は必ずしも全ての転写活性を失っているわけではないことを見いだした(投稿準備中).
3)p53に相互作用し,転写活性を上昇させるc-Abl,p33ING1,ING2を同時導入し,それらによる転写活性の上昇を確認した.
4)Tcf-4/β-cateninの転写活性を測定するアッセイも同様に確立し,APCを同時導入して転写活性が抑制されることを確認した(投稿準備中).
さらに本年度中にp53,p73,p63遺伝子またはTcf-4/β-cateninとともにcDNA libraryをADE2またはFirefly luciferaseのリポーター酵母株に導入し,これらの転写活性を調節する新しい遺伝子のクローニングをモデル実験として行う予定である.
