Research Abstract

ADAM3は受精に関与している精子表面に存在する糖タンパクである。遺伝子工学的にADAM3を作製し、その機能を調べたところ、ADAM3は精子-卵子間の結合を阻害することを見い出した。Mutationを導入して解析したところ、disintegrin loopの7番目のグルタミンが非常に重要であることを見い出した。ADAM3を被覆したビーズは卵子に結合ができることを見い出した。このビーズとの結合はfunction-blocking抗インテグリンα6抗体で阻害された。また、CD9,CD81とCD98(FRP-1)が卵子の表面に発現していることを証明した。抗CD9抗体と抗CD98(FRP-1)抗体がin vitroの受精とADAM3 disintegrin domainの結合能を阻止しうることを見い出した。これらの結果から明らかになったことは、精子-卵子間の結合と融合には精子に存在する多数のADAM familyとFRP-1を含む多数の卵子に存在するインテグリンβ1結合タンパクが関与していることである。FRP-1 light chainが受精の初期過程に関与しているかどうかについても現在解析中である。
