Research Abstract

口蓋扁桃摘出手術やアデノイド切除術により得られたヒト扁桃リンパ球を採取し、フローサイトメトリーにより、リンパ球サブセットさらには抗原提示細胞について検討した。その結果、幼小児期から成人に至るまでのBおよびT細胞の割合、T細胞のサブセットであるヘルパーTリンパ球と抑制性Tリンパ球の割合についても詳細に検討し、細菌抗原の抗原提示に関わる抗原提示細胞の分布状況や、抗原認識や抗体産生の調節に関るTリンパ球サブセットの解析を行うことができた。さらに、ヒト扁桃リンパ球の共通粘膜免疫系組織へのホーミングならびに局所粘膜での機能分化について検討するため、ヒト扁桃由来の抗原特異的抗体産生B前駆細胞や抗体産生を調節するT細胞(1x10^8)をNOD-scidマウスに腹腔内投与し、経時的に脾臓に定着するかどうかについて検討した。その結果、ヒト扁桃リンパ球がTリンパ球を中心として、NOD-scidマウスに定着することが、成人より得られたヒト扁桃リンパ球を移入した際のフローサイトメトリーの結果より明らかにされた。
さらに、定着したNOD-scidマウスにおいて、β溶連菌由来のM蛋白を粘膜アジュバントであるコレラトキシンと共に点鼻感作し、鼻粘膜局所での免疫応答ならびにヒト扁桃リンパ球のホーミングについて検討した。その結果、ヒト扁桃リンパ球を移入したマウス血清中には、M蛋白特異的なヒトIgGおよびIgA抗体が検出され、さらにM蛋白で点鼻感作した移入マウスでは、点鼻感作を行わなかった移入マウスに比べ、鼻洗浄液中のM蛋白特異的ヒトIgA抗体が有意に上昇していた。これらの結果は、ヒト口蓋扁桃リンパ球が共通粘膜免疫系の組織に帰趨し、分化・増殖することにより、粘膜局所で抗原特異的免疫応答を行っていることを示唆していると考えられる。
