Research Abstract

哺乳類の新皮質は哺乳類独自の脳の構造であり、進化の過程で拡大し、複雑化した領域である。また、この脳領域は種々の構能と分かれており、脳の機能の階層化と関与していることが明らかとなっている。本研究ではこの大脳皮質の形成と機能に関わる遺伝子群に注目して新たな分子メカニズムを明らかにすることを目的とする。我々は現在までに、Fyn欠損マウスの作製と解析により、Fynが新皮質の形成に関わっていることを明らかにしてきた。本研究では、このFynを新皮質形成分子と位置付けて、Fynと関連する遺伝子の解析を行い、新たなカドヘリンファミリー(CNRファミリー)との結合を明らかにした。その結果、興味深いことにこのCNRファミリーが、哺乳類の大脳皮質の層構造形成に関わるReelinタンパク質の多重受容体であめることが明らかとなった。また、CNRフアミリーは、ゲノムDNAにおいてB細胞受容体やT細胞受容体に類似した遺伝子クラスターを形成しており、各CNRファミリー間で第一カドベリン領域のアミノ酸配列が高く保存されていた。この第一カドヘリン領域とReelinタンパク質とが結合することが明らかとなり、このReelin結合領域に対する抗CNR抗体によりReelin依存的なDab1のチロシンキン酸化が阻害されることが明らかになった。また更に興味深いことに、この第一カドヘリン領域は動物種の違いにより異なり、魚類では相同な遺伝子の存在が確認できなかったが、両生類では2種類、爬虫類では4種類、鳥類では8種類の相同な遺伝子の存在が想定されており、分子進化が急激におこっている遺伝子である可能性が考えられる。また、ヒトとマウスの遺伝子クラスターの解析より、協調進化がおこっていることが明らかとなっている。脊椎動物の進化の過程で、脳の拡大、複雑化、高次化がおこったことを考えた時、このCNRフアミリーの分子進化は極めて興味深いものであると思われる。今後は、各脊椎動物の遺伝子クラスターの解析を行い、分子進化と脳の進化との関わりあいを明らかにする予定である。
