Research Abstract

1)カリウムイオンによるG-四重鎖DNA形成 : まず、KCI溶液で四分子G-四重鎖の形成の直接可視化と単一分子解祈を行った。塩化カリウムの非存在下では、組み込まれた二重鎖は相互作用がなく、お互いに平行のDNAが観測された。塩化カリウムの添加によりG-四重鎖構造の形成を誘導した。さらに、KCl溶液添加によるG-四重鎖形成と、KC1を含まない緩衝液の添加でG-四重鎖が破壊できることをリアルタイムで観測できた。DNA折り紙を使って我々はストランドの向き、トポロジーおよびG-四重鎖の化学量論を制御すること成功した。
2) G-四重鎖結合性リガンドによって誘導されるDNAのシナプス構造 : 次に、一方の端にリンカーを含むビオチンでbisquinoliniumピリジンジカルボキサミドによって誘発されるG-四重鎖の形成の直接観察を検討した。リガンドの存在下では、特徴的なX字構造が観測されG-四重鎖構造の形成を誘導した。リガンドによって誘導されるG-四重鎖にストレプトアビジンを添加すると、X字状の中央にタンパク質が結合しリガンドが四重鎖に結合していることを示した。
3)分子シャペロンとしてのHIV-1ヌクレオカプシドタンパク質 : HIV-1ヌクレオカプシドタンパク質は、核酸シャペロンと呼ばれ、これまでのところ化学量論、NCP-NCPの相互作用、G-四重鎖形成にシャペロン活性についてはあまりよく分かっていない。我々は、DNA折り紙を使って前駆体タンパク質NCPl5、成熟NCp7の性質について直接的およびリアルタイム分析を行った。タンパク質粒子は、主に溶液中では単量体の形態で存在することが見出され、二量体及び多量体は、溶液中および、四重鎖構造に結合状態で観察された。G-四重鎖への結合形成や解離の様子がリアルタイムに観察され、中間体の状態もまた可視化した。
