Research Abstract

申請者は前年度までに、TLRシグナルの負の制御因子であるTANKがTRAF6のユビキチン化抑制を介した破骨細胞分化の負の制御因子であることを見出した(Maruyama et al. J. Biol. Chem)。また、転写因子」dp2が破骨細胞特異的遺伝子の誘導と好中球の正常分化に必須であることを明らかにした(Maruyama et al. Immunity)。本年度は計画の最終年度のため、「自然免疫系に関与せずに」骨代謝系を制御しているが故に解析が後回しとなっていた遺伝子群の解析に着手した。以下、個々の研究成果を概説する。
1. 転写調節因子Sbno2の骨代謝における機能解析
元来Sbno2はNF-kBを抑制することが報告されていた。そこでSbno2欠損マウスを作成したところ、Sbno2は炎症応答には関与していないことが判明した。一方で、Sbno2欠損マウスは大理石骨病を発症し、破骨細胞融合の障害が観察された。Sbno2欠損破骨細胞ではDC-STAMPの発現が減弱しており、DC-STAMPの導入はその融合障害を救済した。さらに、Sbno2はDC-STAMPプロモーターを抑制する転写因子であるTallと結合することでその活性を阻害していた。以上より、Sbno2による破骨細胞融合の促進を介した骨量制御機構が明らかとなった(Maruyama et al. JEM本報告はJEMの表紙を飾った)。
2. 液性因子Xの骨代謝における機能解析
申請者は分子Xが骨芽細胞及び滑膜細胞において発現しており、この分子が破骨細胞の分化に影響を与えることなく融合だけを抑制することを発見した。分子XはIL-17刺激によって誘導され、RA患者の関節液中において高い濃度で検出される。興味深いことに、OA患者における分子Xの関節液中濃度と骨破壊マーカー・濃度は逆相関した。そこで分子Xを関節リウマチマウスモデルへ投与したところ、骨破壊を抑制できた。さらに、分子Xの欠損マウスを解析したところ、海綿骨量の減少が認められた。現在、分子Xによる破骨細胞融合抑制機構の探究を行っており、成果の一部については特許出顧の上(丸山健太ら、特願2013-235205)国際誌に投稿中である(Maruyarna et al, corresponding author, under revision)。
尚、上記研究成果により第6回Merck award for young biochemistry researcher最優秀賞および井上研究奨励賞を受賞した。

筆頭著者・責任著者での論文報告を計画通りに遂行できている。Human immunologyの導入展開も着実に前進している。

Strategy for Future Research Activity

マウス管理・消耗品購入のための資金が不足しているため、これまで以上に積極的な助成金の申請をおこなってゆく。短期間で着実な成果を上げるため、共同研究や受託外注サービスなどを駆使することで徹底したデータ取得の効率化と合理化を図ってゆく。
