Research Abstract

本年度は"Candidatus Brocadia sinica"のプロテオーム解析のデータについて詳細な解析を行い、細胞内代謝マップの作成を行った。代謝マップ解析ではKEGG、Breda等のデータベースを用いて解析を行い、文献レビューを加えることで各代謝経路の成り立ちを考察した。主要な代謝経路として、嫌気性アンモニア酸化(anammox)、無機炭酸の固定に利用される還元的acetyl CoA経路、還元力生成のための亜硝酸酸化経路を同定することに成功した。特にanammox経路に関与するヒドラジン酸化酵素とヒドラジン合成酵素の発現量が"Ca. Brocadia sinica"の細胞内で極めて高く、主たるエネルギー生産経路となっていたことが明らかとなった。さらに、NrfAの発現がプロテオームから確認されており、これは"Ca. Brocadia sinica"が異化的硝酸還元を行っていた事を示唆する結果だった。"Ca. Brocadia sinica"による異化的硝酸還元を実証するため、細胞に硝酸とギ酸を添加し、嫌気的に培養し、亜硝酸とアンモニアの生成を同位体比分析で調査した。その結果、亜硝酸とアンモニアの生成を確認することができ、"Ca. Brocadia sinica"の新たな生理学的特性を明らかにすることに成功した。一方、興味深いことに亜硝酸還元を担う酵素がゲノム上からも見いだされなかった。そこで、亜硝酸還元酵素を同定すべくRadboud大学(オランダ)に留学し、タンパク質精製の研究に取り組んだ。細胞粗抽出液から亜硝酸還元活性を示すフラクションを確認し、段階的に精製することに成功した。一連の解析を通じて、"Ca. Brocadia sinica"の新たな生理学的特性を明らかにすることに成功したと言える。こうした解析の経過について論文誌において報告を行った。
