Outline of Annual Research Achievements

環境汚染物質の胎児期および乳児期の曝露は出生児の体重あるいは発達に影響することが報告される。本課題では、その影響に関して子宮内発育遅延で関連が指摘されるInsulin-like growth factor 1 (IGF1)に着目し、環境汚染物質との関連を検討することを目的としている.
東北コホート調査(Tohoku Study of Child Development, TSCD)の対象者のうち、曝露指標となる臍帯血中総水銀、ヒ素、カドミウム、セレン、スズおよび鉛ならびにポリ塩化ビフェニル(PCB)濃度データが揃い、かつ母体血の採取時期および出産時年齢等を考慮した約100検体を分析対象とした。IGF1およびInsulin-like growth factor binding protein (IGFBP1)の分析はELISA法により行った。出生時体重、IGF1および環境汚染物質との関連性は、ピアソンの積率相関係数および重回帰分析により解析し、得られた結果を検証するため、構造方程式モデリングによる解析も行った。統計解析はJMP ver.9およびSPSS Amos ver.22によった。
その結果、母体血および臍帯血中IGF-1は出生児の身長および体重と正に相関した。臍帯血中IGF-1と臍帯血中総PCB、3、4、5、6および7塩素体との間に負の関連性がみられた。一方で各重金属濃度との関連性は観察されなかった。また、臍帯血中IGF-1を目的変数、臍帯血中PCB濃度および交絡要因を独立変数とした重回帰分析を実施したところ、臍帯血中IGF-1と臍帯血中総PCBの間にも負の関連がみられたことから、これらの結果をもとに構造方程式モデリング分析を行ったところ、PCB曝露の増加に伴い臍帯血中IGF-1が低下し、出生時体重に影響している可能性が示唆された。臍帯血中PCBレベルとIGF-1との関連についてのヒトでの報告はこれまでなく、この新規知見を国内学会等で発表した。
