Research Abstract

これまでの研究成果より、イトヨにおける日長応答性とその喪失に関与する新しい候補因子として甲状腺刺激ホルモン(TSHβ2)が見出された。海型と淡水型の脳におけるトランスクリプトーム解析を行うと海型では短日から長日への移行時に脳のTSHβ2の発現量が急激に下がる一方、淡水型ではこの日長応答性が失われていた。このTSHβ2の日長応答性の喪失は、独立に進化したふたつの淡水型(北米/日本)で共に見られた。これら、TSHβ2の日長応答性の喪失の遺伝基盤を解明するために、北米と日本の海型と淡水型でF1交雑個体を作成し、TSHβ2の発現解析をすると、TSHβ2の日長応答性の喪失は北米と日本で異なる遺伝基盤を持つことが示された。またこれらのF1交雑個体を用いたアリル特異的発現解析より、北米ではTSHβ2のcis制御領域の違いが、日本ではcis制御領域とtrans因子の両方の違いがTSHβ2の日長応答性の喪失をもたらしていると示唆された。そこで、その詳細な遺伝基盤を探るために、北米の淡水型に注目し、TSHβ2上流配列のSNP/挿入欠失変異の解析を行ったところ、上流1000bp以内に、転写因子STAT5B-like結合部位を含む淡水型特異的欠失領域が発見された。更に、TSHβ2に日長情報をもたらす転写因子を探索するために、北米の海型を用いて、日長変化前と、日超変化後1日の下垂体を用いたRNAseq解析を行うと、このSTAT5B-likeを含むいくつかの転写因子の発現が日長変化後1日以内に変動することが分かった。以上のことから、北米の淡水型では、日長に応じて発現変動をする転写因子STAT5B-likeの結合部位が上流配列から欠失したことで、TSHβ2の日長応答性が失われていると示唆される。このため、現在、この欠失領域のプロモーター解析を行っている。更に、TSHβ2の機能解析のため、TALEN法を用いたノックアウトイトヨを作成し終え、現在F1個体を得ようとしている。
