単為生殖型肝蛭の起源解明に関する研究

Research Abstract

アジアには減数分裂異常により精子形成能を失った結果、単為生殖により増殖すると考えられている単為生殖型肝蛭(Aspermic Fasciolafluke)が分布する。我々は、単為生殖型肝蛭は確定的な種としてよく知られているF. hepaticaとF. giganticaの種間交雑により中国において出現したことを明らかにしてきた。そこで、本年度は中国で誕生した単為生殖型肝蛭がどのようにして周辺諸国へ分布を拡大したかを解明することを目的として研究を行った。
中国と地理的・歴史的関係が非常に深い日本および韓国に着目した。日本および韓国には単為生殖型肝蛭のみが分布すると報告されてきたが、両者の系統関係に関する分子学的解析は不足していた。そこで、日本産肝蛭60虫体および韓国産肝蛭33虫体についてFasciola属の系統解析に最もよく用いられるミトコンドリアDNAのNADH dehydrogenase1(nad1)領域の塩基配列(535bp)に加えて、cytochrome c oxidase 1(cox1)領域の塩基配列(438bp)の解析を行った。その結果、両領域の連結塩基配列に基づき3つのハプロタイプが識別され、そのうち2つのハプロタイプが日本産および韓国産肝蛭で共通であった。したがって、日本産肝蛭と韓国産肝蛭は連続性のある個体群であり、共通の母系祖先を有することが示された。また、日本産および韓国産肝蛭で共通して確認された2つのnad1ハプロタイプの塩基配列は中国産単為生殖型肝蛭で確認されたハプロタイプの塩基配列とそれぞれ完全に一致したことから、日本および韓国に分布する単為生殖型肝蛭は中国を起源とする系統であることが明らかになった。肝蛭の主な終宿主は反鯛類である。日本の在来牛の祖先となったウシは2世紀頃に朝鮮半島から導入されたと考えられていることから、中国で誕生した単為生殖型肝蛭は終宿主であるウシとともに韓国を経由して人為的に日本へ移入したと考えられた。
