Research Abstract

植物ウイルスは化学農薬による防除が不可能なため、近縁の品種や野生種との交配による抵抗性育種が主要な防除手段である。しかし、交配による育種は開発期間が長期にわたる上に、利用できる遺伝的リソースが交配可能な種に限られているため、ウイルスの素早く多様な変異に対応することができず、たびたび打破されてしまう。こうした状況の下、より強力で持続性のある新規ウイルス抵抗性戦略が希求されている。本研究では、非宿主植物からウイルスの複製を強力に抑制する「非宿主抵抗性因子」を単離することにより、全く新しい形の植物ウイルスの高度防除戦略を構築することを目的とする。
本年度は、昨年度同定した、植物ウイルスに対する非宿主抵抗性因子を実際に宿主であるナス科植物に導入し、「非宿主化」することを目指した。まず、非宿主抵抗性因子をバイナリベクター中の35Sプロモーター下流に組み込んだ。続いて、アグロバクテリウムに形質転換し、ナス科植物の葉と共培養し、除菌後に組織培養をおこない、カルス化、再分化を経て再生植物個体を得た。実際に非宿主抵抗性因子を発現していることを確認後、この植物にGFP発現ウイルスを接種した。その結果、ウイルス増殖の指標であるGFP蛍光が全く観察されず、ウイルス増殖が強く抑制されていることが示唆された。このことから、実際にナス科植物が非宿主化されていることが示された。
また、その他に複数の植物ウイルスについて塩基配列やタンパク質発現様式等の基礎的な性状を解析するとともに、感染性クローンの構築をおこなった。それらウイルスは、上記のような非宿主抵抗性の解析をおこなうためのツールとして利用できるが、海外においてもほとんど解析が進んでいないため、今後も充分に性状を明らかにする必要があると考えられた。
