Research Abstract

本年度は、固形癌の中でも最も悪性度が高い膵癌を対象に、固形癌に特徴的な癌微小環境である低酸素環境とMUC1発現および機能的意義の解明を試み以下の結果を得た。(1)低酸素(1%02)下で培養した膵癌の培養上清には血管内皮細胞(HUVEC)の管腔形成誘導能があること、(2)転移性腫瘍から樹立された膵癌細胞株(AsPC1, Capanlなど)では顕著なMUC1の低酸素応答性の発現上昇がみられ、MUC1-CT (MUC1-cytoplasmic tail)が膜面から核へ移行すること、(3)核へ移行したMUC1-CTはp53やβ-cateninを伴い血管新生因子であるCTGFの転写及び分泌を強く促進すること、(4)低酸素性MUC1発現はさらに他の血管新生因子VEGFAやPDGFBの分泌促進にも寄与することを明らかにした(Kitamoto et al. Oncogene, 2013)。近年Mukherjeeら米研究グループが、MUC1を膵臓特異的にノックアウト/ノックインした膵癌モデルマウスを用いて、MUC1の高発現がVEGF等の血管新生因子の発現亢進を伴う多臓器転移能の獲得に重要な役割を担う分子であることを報告している。今回の知見は、これら報告とも一致する結果であり、膵癌内の低酸素環境がMUC1を介した転移能獲得の鍵となる因子であることを示唆している。
本研究は、MUC1が癌細胞自身の形質だけでなく、癌細胞周囲の低酸素環境を感知し、血管新生を誘導することで膵癌の高い転移能の獲得に寄与する可能性を世界に先駆けて明らかにした研究成果であり、これら知見は、今後MUC1を対象とした臨床応用を目指す上で重要な知見となると考えられる。
