Research Abstract

質量分析法の高性能化に伴い、プロテオームと呼ばれる生命現象をタンパク質からアプローチする学問が急速に発展している。さらにタンパク質の機能はリン酸化や糖鎖付加などの翻訳後修飾により制御されていることが多く、この翻訳後修飾の解析は生命現象を理解する上で重要であると考えられている。しかしながら、質量分析によるタンパク質の解析に広く用いられる衝突誘起解離(CID)法では、分析の際に翻訳後修飾の情報が失われてしまうことが多く、新たな分析法の開発が望まれている。翻訳後修飾の解析にはマトリックス支援レーザー脱離イオン化インソース分解(MALDI-ISD)法および電子移動解離(ETD)法が適しているとされているが、従来のCID法に比べ検出感度が低い点が欠点である。本研究ではこれらの手法の高感度化を目的として、物理化学の観点からアプローチした。
MALDI-ISDは、マトリックスとペプチドまたはタンパク質の混合結晶にレーザーを照射することによるタンパク質分子のラジカル分解である。タンパク質のラジカル化は結晶中で起こり、その後の分解はペプチド分子の脱離後、気相中で起こることを明らかにした。このMALDI-ISDを超高分解能質量分析計および新規ソフトウエアと組み合わせることでタンパク質を容易に同定可能となった。また、MALDI-ISDは通常、ペプチドへの水素ラジカル付加による分解反応であるが、私はペプチドからの水素引抜反応でも分解が起こることを明らかにした。この分解反応は従来のMALDI-ISDと相補的な情報を与えるため、タンパク質の同定をより確実に行える。さらに水素引抜MALDI-ISDは糖鎖の構造解析についても有用であった。
一方、ETDはプロトン化したタンパク質と負イオンの電子移動による分解反応である。ETDはタンパク質分子の分解効率が低いことが問題点とされているが、金属とペプチドの複合体形成を利用することでETDにおける分解効率が向上することを明らかにした。さらに分子力学計算を用いて、ETDプロセスの本質にアプローチした。

Reason

当初は水素ラジカル付加反応を利用したタンパク質の翻訳後修飾解析を行う計画であったが、研究の過程において水素ラジカル引き抜き反応によってもタンパク質の分解が起こることを明らかにした。さらに電子付加による分解反応についても研究を行い、3年間の研究期間において18件の査読付論文を発表した。

Strategy for Future Research Activity

今後は、今回明らかにしたタンパク質の分解プロセスを利用し、より高感度にタンパク質の翻訳後修飾解析を可能とする分析法の開発を目指していく。
