高精度位置決め磁気駆動マイクロデバイスを用いたオンチップ細胞操作に関する研究

Research Abstract

本研究ではマイクロ流体チップ内で非接触に駆動される磁気駆動マイクロツール(Magnetically Driven Microtool:MMT)にかかる摩擦を低減することで,マイクロ流体チップ内において細胞の精密・高速操作を行うことを目的としている.駆動源として永久磁石を用いるため,熱の発生がなく,非接触でmNレベルの力を発生させられる超精密・超高速アクチュエータは世界初である.
そこで本年度はMMTに規則的に配列したV溝群(リブレット)を加工することにより,高速駆動時における流体摩擦の低減を行った.まずリブレット解析モデルを作製し,流体摩擦を最小に抑えるためのリブレット形状の最適化設計を実施.この最適形状を作製については,シリコンとニッケルの複合加工を用いることで,高精度に微小V溝群をMMTに加工することを達成した.作製したMMTの駆動周波数特性を測定すると,従来5Hz程度で駆動できなくなっていたのに対して,リブレット付のMMTにおいては90Hz(282mm/s)の高速駆動を達成した.また,マイクロ流体チップのガラス基板に圧電セラミックスを付与して超音波振動を印加することにより,見かけの摩擦力を大幅に低減することが可能であり,これにより最小1.1μmの位置決め精度を達成している.
マイクロ流体チップの安定した環境内において,流体制御をチップデザインにより行うことで,細胞を連続的にMMT作業エリアに搬送することにより,細胞の処理速度を飛躍的に向上することが可能である,本年度はマイクロ流体チップ内において流体微粒子多分岐ソーティング,細胞の多分岐搬送,卵子の高速除核を達成した.mNの発生力mmの位置決め精度,280mm/s以上の高速駆動を達成した非接触アクチュエータは世界初であり,マルチスケールの細胞に対して適応することが可能である.
