Research Abstract

(1)新規神経栄養因子としてのHGFの機能解析
(1)初代培養神経細胞を用いた各種神経栄養活性の解析を行った。特に血清除去の条件に加え、グルタミン酸誘導性の神経細胞死や細胞移動、増殖活性の有無について解析を加えた。
(2)神経特異的HGF発現トランスジェニックマウス(HGF-Tg)を作成した。
(3)運動ニューロン疾患である筋萎縮性側索硬化症(ALS)の進行過程におけるHGF,およびc-Metの発現調節を、SOD1G93A変異をもつALSモデルトランスジェニックマウス(ALS-Tg)を用いて詳細に解析した。
(4)新規神経栄養因子としてのHGFのALSにおける機能を、HGF-TgとALS-Tgのダブルトランスジェニックマウスを作成し、解析した。その結果HGF遺伝子をALS-Tgの神経細胞へ直接長期発現することでALS-Tgの病態進行を遅延させ延命効果を発揮することが明らかとなった。
(2)HGFファミリー分子(HLP)の新規神経栄養因子としての同定と発現解析
(1)HLPは微量の他の神経栄養因子共存下で鶏胚の末梢神経節の神経突起伸長と細胞移動を促進した。この作用はHLPの受容体Ronの細胞外ドメインとIgG-Fcとの融合蛋白(Ron-Fc)で阻害されたことから、HLPと微量の他の神経栄養因子による神経栄養活性はRon/HLP受容体シグナルに依存することが明らかとなった。
(2)マウス末梢神経節におけるHGF,HLP,c-Met,Ronの発生過程における発現解析を行った。
(3)新規スカベンジャー受容体Scavenger Receptor with C-type Lectin(SRCL)のクローニングと機能解析
Yeast two-hybrid法で得られた部分断片をもとに、新規スカベンジャー受容体SRCLをクローニングした。SRCLは2型の膜蛋白質で、SR-A等のスカベンジャー受容体に見られるcollagen-likeドメインに加え,C-type Lectinドメインをもつことを特徴とする。CHO-K1細胞にSRCLを発現させると、細菌(E.ColiやS.aureus)への結合能を獲得した。脳や培養ミクログリアにも発現を認めることから神経系における機能が示唆された。
