Research Abstract

1)分子力学計算により存在を明らかにした種々の超二次構造の安定化を計るため、その素構造であるα-ヘアピン構造とβ-ヘアピン構造について、静電相互作用やジスルフィド結合の適切な導入位置の検討を試みた。Lys残基-Glu残基対の導入によるα-ヘアピン構造の安定化の場合、検討を行った66の導入位置の内、37位置が残基対の導入位置として望ましいことが明らかになった。最大で1.3kcal/molのLys残基-Glu残基対導入による安定化効果が得られた。ジスルフィド結合の導入による安定化の場合、検討を行った66の導入位置の内、7位置のみが残基対の導入位置として望ましいことが明らかになった。共有結合形成という安定化効果の優位さが、逆に導入位置の制約を大きくしており、分子力学計算による導入位置予測を、実験に先立って行うことの重要性を示唆する結果となった。ジスルフィド結合の導入によるβ-ヘアピン構造の安定化の場合についても、導入位置の制限が大きいことが示され、分子力学計算による導入位置予測の有効性が明らかになった。2)プロリンを主成分とするポリペプチドを縮合法で合成し、水溶液の温度変化による凝集転移挙動を検討した。副成分のアミノ酸の疎水性の大きさと、ポリプロリン-II構造の局所的な乱れの大きさに対応して、転移温度が大きく変化することを見いだした。このことは、副成分として用いるアミノ酸の疎水性とコンホメーション特性に着目することにより、任意の転移温度を有する温度応答性ポリペプチドを分子設計しうる可能性を示しており、薬物徐放用分子としての展開という視点において興味ある現象といえる。
