Research Abstract

1.Fasを介するアポトーシス誘導シグナルが、Ras/ERKの活性化と、NF-kBの活性化による異なったメカニズムで阻害されることを示した。また、自己免疫疾患や成人T細胞白血病を発症させるHTLV-1のがん遺伝子産物Taxがc-FLIPの発現を強く誘導することによって、Fasを介するアポトーシスを抑制することを示した。
2.マクロファージに発現する新規スカベンジャーレセプターSR-PSOXをクローニングしたが、この分子が活性化T細胞やNKT細胞に発現すそケモカインレセプターCXCR6のリガンドである膜結合型ケモカインCXCL16と同一分子であり、スカベンジャーレセプター活性とケモカイン活性という異なった二つの活性を有する新規分子であることを明かとした。また、SR-PSOXがDCによる細菌の貪食に機能することを中和モノクローナル抗体を用いた解析で明らかにした。
3.CD40シグナルにおけるTRAF6の役割を解析し、CD40シグナルによるNFκB活性化にTRAF6が必須であることを証明した。また、野生型マウスとTRAF6欠損マウスを比較し、TRAF6欠損マウスにおいてRANK-TRAF6シグナルによるLTa1b2の産生が消失するためリンパ節原基の形成が不全となることを明らかとした。次に、TRAF6欠損線維芽細胞に種々のTRAF6変異体を発現させ、IL-1やLPS刺激依存性のNFκB及びMAPKの活性化を解析し、TRAF6の機能部位を同定した。そして、TRAF6N末端Zn結合領域中に三つの区別されうる機能部位が存在することを明らかにした。またTRAF6結合分子、TIFAを同定した。TIFAはTRAF6と恒常的に結合しているのに対してIRAK-1とはIL-1刺激依存的に結合した。即ちTIFAはIL-1シグナルにおけるTRAF6とIRAK-1のリンカータンパク質であることを示した。
