Research Abstract

私どもはこの結合部位の3次構造を認識するモノクロナール抗体と、ランダムペプチドを提示するファージライブラリーを用いて、リセプターとリガンドの結合ドメインミミックを直接デザインできるかという構想について、検討してきた(Nature biotech.,16:267-270,1998,J.Immunol.,161:6622-6628,1998,Mol.Immunol.36,1249-1254,1999,J.Biochem.in press)
MCP-1では:sC27とsG25と呼ぶMCP-1のケモタキシス活性を阻害するペプチドモチーフの合成に成功した(J.Biochem.in press)。
IL-5では:IL-5とIL-5Raとの結合を阻害するTB-13抗体を用い、2種類のペプチドモチーフを明らかにしたが、これらのファージクローンにはIL-5依存性のTF-1細胞の増殖への阻害活性が認められなかった(Proc.Jap.Soc.Immunol.,29:pp210,1999)。ヒト細胞系によるIL-5活性の評価システムを検討する必要がある。
IL-6では:ファージライブラリー(PhDC7C:BioLabs)を用い、抗IL-6抗体(B-E8:Diaclone)が特異的に認識するモチーフを5種類明らかにした。これらのファージクローンはヒトIL-6に依存して増殖するヒト細胞株、KT-3の増殖を阻止するが、ヒトIL-6に反応性のマウス細胞MH60の増殖を阻害しないという興味深い阻害活性を明らかにした。
