Research Abstract

嗅球での解析を主として進めた。ギャップ結合・シナプス等のニューロン相互関係の電子顕微鏡レベルでの形態学的解析を進め、特に、糸球体特異的な同期活動に対応するギャップ結合をより明確に示した。海馬、大脳皮質ではギャップ結合は主に同一タイプのニューロン間に見られる。しかし、ギャップ結合は決して同種のニューロン間だけに限られるわけではない。嗅球におけるギャップ結合の解析の結果、従来考えられていたこととは異なり,ギャップ結合は多様なニューロン間で形成されていることを示した。特に,糸球体では、我々が発見し、提唱したtype1,2の傍糸球体細胞のうち、嗅上皮感覚ニューロン軸策終末からシナプスを受けているtype1傍糸球体細胞が僧帽細胞・房飾細胞の一次樹状突起tuftとの間でギャップ結合を作っていることを明らかにした。しかし、type1傍糸球体細胞の中心グループであるGABA陽性およびドーパミン性傍糸球体細胞も、ギャップ結合を形成している主要なニューロンではなく、まだ同定されていない別のtype1傍糸球体細胞の存在が示唆された。更に、傍糸球体細胞以外にギャップ結合を投射ニューロンと形成する新たな介在ニューロンの存在も示唆した。一方、詳細な電子顕微鏡レベルでの連続切片-再構築法により、投射ニューロン間にもギャップ結合が存在することを形態学的に始めて証明した。これらデータに基づいて,最近注目され始めた糸球体特異的な同期したニューロン活動の機構として、投射ニューロン間のギャップ結合による直接の相互作用及び、介在ニューロンとのギャップ結合または混合シナプス(ギャップ結合+化学シナプス)による間接的な相互作用の双方を可能性として示唆した。
