Research Abstract

学習行動下での脳内情報処理過程を明らかにする為に、
1)櫻井芳雄京都大学教授との共同研究で、AVD(Audio Visual Discrimination)-task下でのc-Fos遺伝子発現を調べているが、この課題では、音は、高低、光では左右という音と光以外にも異なるパラーメーターが入っていた(Sakata et al.,Eur J Neurosci,15,735-743,2001)。そこで、同じ位置の白色光と白色音刺激に対して、nose poke反応するよう装置を改良した。このシステムを用いて、音から光に刺激を転換した後にも位置情報の空間記憶が残っていること、又、光と音を同時に与えた時に、反応時間が早くなる事を確認した(Sakata et al.,Exp Brain Res.2004)。
2)Wheel running装置を考案した。このシステムでは、24時間絶飲させたマウスを回転ホイール装置に入れる。一定方向にホイールが回転する為、給水口から出る水を効率良く飲む為には、マウスはホイール内で足場となるペグ上をそのパターンに合わせて走行しなければならない。走行パターンを変えた直後では、踏み外しの回数が増えるが、1週間でほぼ誤りなく走ることができるようになる。走行パターン変化に伴う脳内に於けるc-Fos発現の変化を調べた。その結果、大脳皮質運動野、線条体、小脳で有為なc-Fos発現の増加が見られた。特に、線条体で最も顕著であった。そこで、線条体の神経細胞種特異的なマーカーとの2重染色法によって、走行パターンの変化とc-Fos発現の関係を調べた。その結果、走行パターンを変えた直後に、nNos(nitric oxide synthetase)陽性細胞でのc-Fosの発現が一過的に増加することが判った。他方、直接経路(direct pathway)や間接経路(indirect pathway)のマーカーである、サブスタンスPやエンケファリン陽性細胞、また介在神経のParvalbumin陽性細胞では、走行中は、顕著にc-Fos発現が増加するものの、走行パターンを学習したあともその発現は増加したままである事が判った。これらの結果は、wheel running中に活動が促進する線条体の神経細胞の中は、パターン変化に特異的なものと、走行そのものに特異的なものの2種類がある可能性を示唆する(Kitsukawa 2002,2004 北米神経科学大会)。
