Research Abstract

1)神経活動依存的に発現する接着分子arcadlin遺伝子のノックアウト(KO)マウスが既に作製済みである。このマウスを用いて、扁桃体連日刺激によるキンドリングの発達過程を解析したところ、ノックアウトマウスの方が脳波上も、また行動学的にもキンドリング形成が早いことが分かった。さらに、キンドリング終了後、Timm染色によって海馬顆粒細胞軸索(苔状線維)のterminal sproutingを解析したところ、ノックアウトマウスの方が発芽が亢進していることも確認された。以上の結果から、Arcadlin分子は異常可塑性の進行を抑制していると考えられた。
2)ArcadlinはN-cadherinと相互作用することを明らかにしている。相互作用する領域を決定するため、N-cadherinの細胞外および細胞内領域欠失変異体をそれぞれ作成したが、いずれもArcadlinとの結合活性は残存していた。そこで、膜貫通領域の561番目のロイシンをプロリンに変え、ヘリックス構造を乱すと、Arcadlinとの結合が減弱することが分かった。このことは、N-cadherinとArcadlinは細胞膜内で結合する可能性を示唆している。
3)Arcadlin細胞外領域の同種結合によって、TAO2→MKK3/6→p38MAPKのsequential phosphorylationとArcadlinのエンドサイトーシスが生じることを明らかにしている。そこで、この現象がニューロン内で起きるかどうかを確かめるため、海馬の培養ニューロンにarcadlin-EGFPを遺伝子導入し、Arcadlin細胞外領域蛋白質を加えたところ、Arcadlin-EGFPがスパイン頭部から樹状突起の方へ移動する像が観察された。
4)動物個体のin vivo imagingのために、まず脳に深く刺入できるレンズを探索した。GRINレンズが、その目的に適していると考え、共焦点顕微鏡と組み合わせて使えるかどうか検討したが、色収差が予想以上に大きいため、難しいことが分かった。二光子レーザー顕微鏡との組み合わせは今後の課題である。
