Research Abstract

これまで、われわれは心筋梗塞において骨髄幹細胞が梗塞部位に動員されることを報告した。しかし、サイトカインを用いて心筋梗塞部位に骨髄幹細胞が誘導され心機能が改善されるか否かは明らかではなかった。そこでえ本研究では、サイトカインによって循環血液中に動員した骨髄幹細胞による組織再生をマウスを用いた骨髄移植の系で評価した。[方法]Lac-ZあるいはGFPトランスジェニックマウスの骨髄細胞を致死量放射線照射後の同系(C57BL/60マウス)に移植した。次いでドナー骨髄細胞の生着が確認されたレシピエントマウスの左冠動脈を、全身麻酔下で結紮することによって心筋梗塞を作成した後、さまざまなサイトカインの連日投与を施行した。その後、心エコーにて心機能を評価したのち心臓を摘出し、免疫組織科学的に骨髄由来細胞による心筋組織再生について検討した。[結果]G-CSF投与群はサイトカイン非投与群に比べて心筋梗塞後の死亡率が低く、リモデリングによる心拡大の抑制および心機能の改善を認めた。さらに、ドナー骨髄由来細胞による心筋細胞、血管内皮細胞、平滑筋細胞の再生が認められた。M-CSFおよびGM-CSF投与群では炎症性細胞の浸潤が強くコントロール群と比べて心拡大および心機能の低下を認めた。G-CSF投与による心筋組織再生は、心筋梗塞の新しい再生治療となる可能性があると考えられた。
