Research Abstract

本研究は全身性またh局所性に骨形成を促進する技術の創生し、骨粗霧症治療法や骨再生技術を開発することを目的としている。またその技術の臨床応用にための関連研究をおこなった。
1.PDEの骨形成促進効果の発見:サイクリックAMP (cAMP)の分解酵素であるphosphodiesterase (PDE)の非選択的阻害剤であるpentxifyline (PTX)の全身連続投与によって骨形成が亢進し、全身の骨量が増加することを明らかにした(Bone,27:2000.)その後、その骨形成促進効果はBMPの骨芽細胞分化反応の促進によることが示された(Bone31:396-401,2002)またこのPTXの作用はPDE4の選択的阻害剤であるrolipramによって再現されたため、多くのPDE isomersのうちPDE4が関与していることを明らかにした(Bone.30:589-93.2002,J Bone Miner Res.17:249-56.2002)。
2.骨損傷(骨折)修復初期でのBMPおよびBMP拮抗蛋白(noggin)遺伝子発現の亢進反応の発見:骨折修復のための仮骨形成に先立ってBMPとnogginの発現亢進がおこり、仮骨形成の制御が行われることを明かにした(J. Bone and Mineral Research,16:2001.).
3.BMPの新規薬物伝達系の開発:従来BMPによる局所的骨形成誘発のための担体として動物由来コラーゲンが用いられてきた(BSEのリスク)。将来の臨床利用を考慮して安全な合成担体として、ポリ乳酸ポリエチレングリコール共重合体ポリマーを開発し、その有効性を証明した(Nature Biotechnology,19;2001)。このポリマーをBMPの担体として用いることについて、特許を得た。
