Research Abstract

ゼブラフィッシュ初期発生系において中性セラミダーゼ遺伝子をノックダウンすると血管新生が著しく遅滞することを見いだした(J.Biol.Chem. 279,44012-44022,2004)。興味深いことに、この血管新生不全は、受精卵に中性セラミダーゼmRNA、スフィンゴシンあるいはスフィンゴシン-1-リン酸(S1P)をマイクロインジェクションすることによりほぼ完全に回復した。一方、S1Pの受容体であるS1P_1をノックダウンすることによっても中性セラミダーゼノックダウン胚と同様に血管新生の不全が惹起されたが、S1Pのマイクロインジェクションでは回復しなかった。以上の結果は、ゼブラフィッシュの初期発生において、中性セラミダーゼはS1Pシグナリングを介して、血管新生に深く関わっていることを示唆している。一方、形質膜表面の中性セラミダーゼが形質膜のセラミドを分解するのか、生成したスフィンゴシンは細胞外でS1Pに変換されるのか、については明確な答えが得られていなかった。今回、HEK293細胞の中性セラミダーゼをRNAiでノックダウンする実験、スフィンゴシンキナーゼを過剰発現する実験等を行い、形質膜セラミドを起点とするS1P生成経路の存在を分子レベルで証明した。また、最終的に生成したS1PがEdg受容体の機能的なリガンドになることを示した。
