Research Abstract

本研究計画に関して、本年度は高等植物(シロイヌナズナ)の時計分子機構に関して詳細な研究を行った。我々は既に時計の中心振動体を形成する一連の遺伝子群(PRR1/PRR3/PRR5/PRR7/PRR9)を発見し、その機能解析を主に遺伝学的に方法で行ってきた。本年度は、PRR7/PRR5、PRR9/PRR7、PRR9/PRR5、及びPRR9/PRR7/PRR5からなる二重及び三重遺伝子欠損植物体を作出して時計関連の表現型を詳細に解析した。その結果、PRR9/PRR7二重遺伝子欠損植物体は長周期(時計が一日30時間で動く)、一方、PRR7/PRR5二重遺伝子欠損植物体は単周期(時計が一日19時間で動く)という両極端の性質を示すことが明らかになった。次いで、PRR9/PRR7/PRR5の三重遺伝子欠損体を作成したところ、この植物がもつ時計はほぼ動かなくなってしまった。このことは、これらの遺伝子か時計の進行スピードを制御する中心振動体の一部(部品)をコードしていることを強く示唆している。これらの結果をもとに、我々は高等植物の生物時計の分子機構に関して新しいモデルを提唱した。また、これらの時計欠損植物体では、開花時期(どの季節に花が咲くか)の制御にも欠陥が生じ、開花時期が大幅に遅延した。また、一般に高等植物は赤色の光に敏感に反応して自らの背丈の伸張や形態を変化させるが(環境への適応)、変異体植物では赤色の光に対する感受性が大きく損なわれていた。このようなことから、本研究計画で我々が解析を進めてきたPRR遺伝子群は、高等植物の高次機能の一つである生物時計(花時計)やそれと関連した開花・結実時期の制御(花時計)、及び光形態形成に深く関わっていることが明らかとなった。また、我々はこれらモデル植物(シロイヌナズナ)に加えて有用穀物(イネ)のPRR時計遺伝子群の解析も平行して行った。その結果、イネにも同様の働きをする時計遺伝子群(OsPRR)が存在することを見いだした。従って、今回我々が提唱した新しい時計分子モデルは色々な高等植物に広く適用できると考えられ、植物バイオサイエンスにおける応用・展開研究に関しても重要な知識基盤となると考えられる。
