Research Abstract

1.窒素栄養に応答したシロイヌナズナのサイトカイニン(CK)合成系遺伝子(AtIPTs)の発現解析から、ATIPT3が窒素栄養に応答した短期的なCK合成に、またAtIPT5より長期間の窒素栄養変動に対するCK応答に関わっていることを明らかにした。
2.イソペンテニル型CKからトランスゼアチン型のCKへの変換を触媒するP450酵素遺伝子(CYP735A)をシロイヌナズナから同定するとともに、生化学的解析を用いて、この水変換反応は従来の予想に反し、ヌクレオチド型CKを基質にして起こることを明らかにした。また、この酵素遺伝子の発現はオーキシンによって抑制されることを見いだし、トランスゼアチン生合成制御におけるオーキシンの役割を明確にした。
3.His・Aspリン酸リレー系の下流で転写制御を受ける遺伝子を探索するため、シロイヌナズナのARR21およびARR22の過剰発現体と野生型間での遺伝子発現プロファイルをマイクロアレイ解析により行い、この情報伝達系の下で制御される遺伝子候補の絞り込みに成功した(名古屋大水野教授との共同研究)。
4.トウモロコシのHis・Aspリン酸リレー系を介したCK情報伝達においてシグナルの統合及び分岐点に当たるZmHPタンパク質の立体構造を明らかにし、相互作用の特異性に関わるアミノ酸残基の推定を行った。
5.トウモロコシのCK配糖化に関わる酵素cis-zeatin-O-glucosyltransferase (cisZOG1)がイソプレノイド型CKのみならず芳香族型CKの修飾にも関わることを明らかにするとともに、cisZOG1のシスゼアチン、トランスゼアチンに対する基質親和性と、CK受容体(ZmHK1)のこれらリガンドに対する親和性の類似性を見いだし、cisZOG1とZmHK1の間での基質(リガンド)認識機構の共通性を指摘した。
