Research Abstract

大腸菌におけるHis・Aspリン酸リレー情報伝達分子機構の解明を目的として、病原性大腸菌O-157特異的ゲノム解析を基にした二成分制御系による病原性遺伝子群解析と、リン酸レギュロンの二成分制御系による遺伝子発現ネットワーク解析により以下のことを明らかにした。
腸管出血性大腸菌O-157のゲノム長は5.6M塩基対でK12株と比べ、O-157特異的領域は約1.6Mである。この特異的領域に存在する二成分制御系をホモロジーに基づくコンピュータによる検索と解析をおこなった結果、K12株にはない二組のO-157特異的二成分制御系を発見して、これらの二成分制御系により発現が制御される遺伝子領域を詳細に明らかにした。さらに、二成分制御系のリン酸レギュロンにおけるモデュレーターPhoUのドミナントネガテブなphoU変異体を用いて、モデュレーターPhoUがおこなうリン酸レギュロンの負の調節機構に関して、PhoU側の機能部位を特定した。
