Research Abstract

活性酸素で生じるDNA損傷に塩基損傷と単鎖切断があるが、特に後者は複製を経て二重鎖切断をもたらしゲノム安定性に関わる重要な損傷であるが、これまで余り研究されてこなかった。その理由の一つは細胞内に単鎖切断のみを作る方法がなく、塩基損傷との混合を解析していたからである。我々はその状況を打開するために、ヒト細胞での単鎖切断の修復を解析する実験系(XPA-UVDEの系)を開発した。この系は、紫外線損傷を修復出来ないヌクレオチド除去修復の欠損ヒトXPA細胞に種々の紫外線損傷の直ぐ5'側にニックを入れるアカパンカビ由来その酵素UVDEを発現させ、紫外線損傷の場所にニックを入れてその修復プロセスを解析する実験方法である。さらに紫外線をフィルター上の小孔を通してヒト細胞核の一部に当てることにより、局所的に紫外線損傷を作り、細胞内で発現させたUVDEの働きにより単鎖切断が局所的に作られる系を開発した。この方法により単鎖切断はその近傍にポリADPリボシル化を招き、そこにXRCC1が集積することで修復が開始されることが初めて明らかになった。また、レーザーを用いた種々の活性酸素損傷をヒト細胞の核に局所的に作成する方法の開発とそれを用いた修復プロセスのリアルタイム解析を行い、XRCC1の集積がなければ、PolβやPCNAが損傷部位に集積しないことを明らかにした。すなわち、単鎖切断も塩基損傷の修復と同じようにショートパッチとロングパッチがあり、XRCC1に依存して修復合成の二つの経路の一つが損傷依存的に機能することを実際の細胞の中で明らかにした。このように、これまでに長く課題とされていた疑問に答え、将来の発展的研究を約束する研究の展開があった。
