Research Abstract

ミスマッチを持つオリゴヌクレオチドを用いた遺伝子修復においてMutSヘテロダイマーによるミスマッチ認識機構を解析する。また遺伝子修復におけるミスマッチ修復(MMR)系および他の修復系の関与を明らかにする。ミスマッチ修復系の認識機構を明らかにするだけでなく遺伝子を改変する技術としての開発も目標とする。
1)無細胞遺伝子修復系の確立とミスマッチ修復の関与:変異したlacZalpha遺伝子をcodeするplasmidを、オリゴヌクレオチドおよび細胞抽出液とin vitroでインキュベートし、回収したplasmidのファージの色を解析した。すると、低い頻度(0.09%)ながら変異塩基の正常塩基への修復を見出した。表現型による遺伝子修復アッセイは煩雑で時間がかかるためDNAを用いたアッセイを検討したが、表現型アッセイを上回る方法は開発できなかった。MMRの関与を調べるために、MMRが正常なHL60細胞の代わりに、hMSH2を欠損しているLoVo細胞、あるいはhMLH1が欠損しているHCT116細胞の抽出液を使用したところ修復効率が上昇し、MMRが遺伝子修復に関与していないことを示唆した。
2)細胞を用いた遺伝子修復系の確立とミスマッチ修復の関与:不活化したneomycin^r遺伝子を標的遺伝子としてstableに発現するHeLa細胞を樹立した。1.5 x 10^5 cellにオリゴヌクレオチドをtransfectしたがneomycin^r細胞は得られなかった。一方、同様の実験をMMRが正常なHeLa細胞の代わりに、MMRが異常なHCT116細胞のStable transformantにおいて行うと、neomycin^r細胞が得られ、塩基配列を調べた結果、標的塩基部位で正常塩基への置換が起こっていることがわかった。
3)標的DNAの断片とミスマッチを持つ単鎖オリゴヌクレオチドとの複合体とhMutSalphaの結合:標的DNAの断片とミスマッチを持つ単鎖オリゴヌクレオチドとの複合体に精製したhMutSalphaを加えるとミスマッチ依存性を持つゲルシフトを示すことから、D-loop中のミスマッチはhMutSalphaにより認識されると考えられた。しかし、無細胞および細胞の修復アッセイの結果からMMRは遺伝子修復が起こらないように働いていることが示唆された。
