Research Abstract

本研究では体節の形成と維持の機構を解明することを目的として、そこに関与する遺伝子の探索と機能解析を2つのアプローチにより行ってきた。その一つはゼブラフィッシュの体節前駆細胞に特異的に発現する遺伝子の探索とその機能解析である。我々はこれまでにin situhybridizationにより、ゼブラフィッシュの体節前駆細胞に特異的に発現する遺伝子を数多く同定してきたが、今年度の研究から、その一つであるher13.2遺伝子は体節の周期性の確立に重要な働きをすることが示された。体節の周期性の確立には分節時計と呼ばれるNotchシグナル伝達系と転写因子Hairy(her)から形成される負のフィードバックループが中心的な役割を果たしており、その維持はFgfやWntなどの分泌性シグナルにより行われている。her13.2遺伝子は体節前駆細胞においてFGFシグナルにより発現が誘導され、しかもその機能阻害実験から分節時計の形成に必須であることが明らかになった。さらに、分節時計の心臓部であるHer1タンパク質と結合し分節時計を協調的に制御することが示された。以上の結果から、FGFシグナルによる分節時計の維持のしくみを理解する上で鍵となる分子の実体とその機能が明らかになった。
一方、この研究と平行して、我々は体節の形成に異常をきたす突然変異体を複数系統樹立してきた。我々はそのような突然変異体の中に頭部側数体節の体節境界の維持に異常を呈するものがあることを見い出し、その原因遺伝子の同定を行った結果、IntegrinとFibronectinが体節境界の維持に必要であることを明らかにした。さらに、体節境界に面した細胞の上皮化がその維持には深く関わっていた。以上の結果から、体節の周期性が確立された後に、形態的に明瞭な境界が形成されるプロセスが明らかになった。
